This method consists of membrane filtration, followed by culturing in a chromogenic agar (CCA) and a calculation of the number of target organisms present in the sample.

Place the membrane filter over a E. Coli Coliforms Chromogenic Agar plate. Invert Petri dish and incubate at 36±2 oCduring 21±3 h.

Used for Simultaneous detection of Escherichia coil and total coliforms in water samples.

Conclusion: To obtain the best results with CHROMagar, the medium should be incubated at 35 °C for 48 h in an atmosphere of 5% CO 2. A control C. albicans strain should be inoculated on each medium plate to dif erentiate the color tones of C. albicans and C. dubliniensis.

Place the membrane filter on rehydrated nutrient pad. Incubate the inoculated nutrient. Interpret the results qualitatively by observing the presence or absence of growth and quantitatively by counting the number of colonies on the surface of the membrane filter and calculating CFU/ml.
